Transgenic poplars: expression of chimeric genes using four different constructs

摘要: Leaf or stem explants of a hybrid poplar clone (Populus tremula X Populus alba), sensitive to Agrobacterium tumefaciens, were co-cultivated either by an octopine nopaline disarmed A. tumefaciens modified strain. Transformed shoots readily regenerated from explants. The protocol was improved using the strain C58/pMP90 with binary vector pBI121. This then used test three other vectors. first one, possessing nptII gene fused CaMV 19S promoter, permitted regeneration transformed in presence 50 100 mg/l kanamycin. two vectors carried additional under control 35S promoter double enhancer sequence (CaMV 70). 70 provided consistently higher level expression than promoters both callus and leaf tissues.
