MDR1 gene C3435T polymorphism and cancer risk: a meta-analysis of 34 case–control studies

摘要: P-glycoprotein, the product of MDR1 gene, is a transmembrane active efflux pump for variety environmental toxins and xenobiotics. Epidemiological studies have evaluated association between C3435T polymorphism cancer susceptibility. However, published data are still inconclusive. To derive more precise assessment this relevance, we performed meta-analysis, up to September 2010, 5,196 cases with different types 6,827 controls from 34 case–control studies. Summary odds ratios (ORs) corresponding 95% confidence intervals (CIs) were estimated using fixed- random-effects models when appropriate. The overall results suggested that variant was associated moderately increased risk in all comparison tested (OR = 1.26, CI: 1.06–1.50 TT vs. CC; OR = 1.19, 1.04–1.37 CT OR = 1.15, 1.01–1.32 recessive model; OR = 1.21, 1.06–1.38 domain model, OR = 1.14, 1.04–1.26 allele contrast). In subgroup analysis by types, significant associations found breast (OR = 1.66, 1.24–2.21 OR = 1.44, 1.14–1.82 OR = 1.41, 1.10–1.81 OR = 1.31, 1.13–1.52 contrast) renal (OR = 1.99, 1.37–2.90 OR = 1.74, 1.25–2.42 OR = 1.43, 1.09–1.88 OR = 1.40, 1.17–1.68 no colorectal cancer, gastric acute lymphoblastic leukemia genetic models. ethnicity analysis, among Caucasians under dominant homozygote comparison, versus CC comparison. summary, meta-analysis suggests susceptibility, increasing cancer.
