摘要: Structural biology of membrane proteins is often limited by the first steps in obtaining sufficient yields because native sources are seldom. Heterologous systems like bacteria then commonly employed for protein over-expression. Escherichia coli main bacterial host used. However, overproduction a foreign at non-physiological level usually toxic cells or leads to inclusion body formation. Those effects can be reduced optimizing cell growth conditions, choosing suitable strain and expression vector, finally co-expressing target b-subunit E. adenosine triphosphate (ATP)-synthase, which triggers proliferation intracytoplasmic membranes. This chapter devoted help experimenter appropriate plasmid/bacterial combination amount produced its correct folded state.
