摘要: Two-dimensional gel electrophoresis (2-DE) is currently the best method for separating complex mixtures of proteins, and its use gradually becoming more common in cardiac proteome analysis. A number variations basic 2-DE have emerged, but their usefulness analyzing tissue has not been evaluated. The purpose present study was to systematically evaluate capabilities limitations several techniques proteins from rat heart tissue. Immobilized pH gradient strips various ranges, parameters protein loading staining, subcellular fractionation, detection phosphorylated were studied. results provide guidance analysis other tissues terms selection isoelectric point window accurate quantitation abundance, stabilization technical variation, reduction sample complexity, enrichment low-abundant proteins.
