摘要: The reproducibility of mass spectrometry (MS) data collected using surface enhanced laser desorption/ionization-time flight (SELDI-TOF) has been questioned. This investigation was designed to test the SELDI over time by multiple users and instruments. Five laboratories prepared arrays once every week for six weeks. Spectra were on separate instruments in individual laboratories. Additionally, all produced each rescanned a single instrument one laboratory. Lab-to-lab array-to-array variability alignment parameters larger than attributable running samples during different coefficient variance (CV) spectrum intensity ranged from 25% at baseline, 80% matrix noise region, about 50% exponential drop maximum noise. Before normalization, median CV peak heights 72% reduced 20% after normalization. spectra common instrument, 5% Normalization 18%. With proper processing methods, produce containing large numbers reproducibly located peaks, with consistent heights.
