摘要: BACKGROUND AND OBJECTIVES: Analysis of IgH rearrangements in B-cell malignancies has provided clinical researchers with a wide range information during the last few years. However, only studies have contributed to characterization these features multiple myeloma (MM), and they been focused on analysis expressed allele only. Comparison between non-functional alleles allows further characterizion selection processes which pre-myeloma cells are submitted. DESIGN METHODS: We analyzed cohort 84 untreated MM patients order characterize their functional VDJH DJH rearrangements. The pattern mutations gene segment usage for both types was by polymerase chain reaction sequencing. RESULTS: VH3 VH1 family members were over- under-represented, respectively. VH3-30 VH3-15 segments most frequently used, whereas VH4-34 found or heavily mutated DH2 DH3 over-represented repertoires, while DH1 under-represented productive Finally, DH3-22 DH2-21 be repertoire commonly used less mature malignancies, such as DH6-19 DH3-3, under-represented. INTERPRETATION CONCLUSIONS: Data reported here help identify clonogenic cell post-germinal center B that undergone germinal reaction.
