Vapor detection and discrimination with a panel of odorant receptors

摘要: Olfactory systems have evolved the extraordinary capability to detect and discriminate volatile odorous molecules (odorants) in environment. Fundamentally, this process relies on interaction of odorants their cognate olfactory receptors (ORs) encoded genome. Here, we conducted a cell-based screen using over 800 mouse ORs against seven odorants, resulting identification set high-affinity and/or broadly-tuned ORs. We then test whether heterologously expressed respond odors presented vapor phase by individually expressing 31 measure cAMP responses odor stimulation. Comparison response profiles demonstrates platform is capable discriminating between structural analogs. Lastly, co-expression carboxyl esterase Ces1d mucosa resulted marked changes activation specific odorant-OR combinations. Altogether, these results establish detection discrimination form basis for an OR-based sensor.
