Intramyocardial Injection of Pig Pluripotent Stem Cells Improves Left Ventricular Function and Perfusion: A Study in a Porcine Model of Acute Myocardial Infarction

摘要: Induced pluripotent stem (iPS) cells have the potential to differentiate various types of cardiovascular repair an injured heart. The therapeutic benefits iPS cell based treatment been established in small-animal models myocardial infarction (MI). We hypothesize that porcine (piPS) transplantation may be effective for MI. After a 90-minute occlusion left anterior descending artery model, undifferentiated piPS or PBS were injected into ischemic myocardium. Cardiac function, perfusion and differentiation investigated. One week after delivery, global ventricular ejection fraction (LVEF) significantly decreased both group compared Sham (p<0.05, respectively). Six weeks LVEF improved (56.68% vs. 50.93%, p = 0.04) but was still lower than group. Likewise, regional injection (19.67% 13.67%, 0.02). infarct area smaller (12.04% 15.98% 0.01). PiPS engrafted myocardium can vessel cells, which result increased formation new vessels infarcted Direct intramyocardial decrease size improve function immunosuppressed AMI model.
