摘要: Rearrangement of the BCL-2 gene is molecular consequence t(14;18) chromosomal translocation, which found in approximately 60-90% follicular lymphomas. To investigate ability polymerase chain reaction (PCR) to detect this rearrangement fixed-tissue samples, we studied 48 cases lymphoma using DNA extracted from paired samples fresh-frozen tissue and formalin-fixed, paraffin-embedded tissue. A standard phenol-chloroform extraction method was used for both types Rearrangements major breakpoint region (MBR) minor cluster sequence (MCS) were examined. Three segments human beta-globin also amplified estimate degree degradation samples. PCR fresh-tissue (intact) revealed amplifiable products 29 lymphomas (60%), whereas (degraded) studies positive 24 (50%). MBR detected varied 80 bp > 1.5 kb. Twenty these yielded corresponding DNA, ranging 276 bp. Five four produced MCS that ranged 340 1.2 Four five with no detectable or translocations degraded had greater than 1.0 kb studies. 175-bp segment all samples; a 324 fragment 20 (69%), 676 13 (45%).(ABSTRACT TRUNCATED AT 250 WORDS)
