Pathogenesis of thrombosis: cellular and pharmacogenetic contributions

摘要: Our understanding of thrombosis formation has evolved significantly ever since physician Rudolf Virchow proposed his "triad" theory in 1856. Modern science elucidated the mechanisms stasis, hypercoagulability, and endothelial dysfunction. Today, we have a firm key molecular factors involved coagulation cascade fibrinolytic system, as well underlying genetic influences. This knowledge cellular contributors been translated into diverse pharmaceutical interventions. Here, examine its associated pathologies. We also review current state pharmacologic interventions, including pro- anti-thrombotics, direct oral anticoagulants, anti-platelet therapies. The pharmacogenetic that guide clinical decision making prognosis are described detail. Finally, explore new approaches to drug discovery, repurposing, diagnostics. argue network biology tools will enable systems pharmacology revolution next generation facilitating precision medicine applications ultimately leading improved patient outcomes.
