Chestnut allergen detection in complex food products: Development and validation of a real-time PCR method

摘要: Abstract Chestnut is gaining importance as food allergen, but a specific detection method has not been performed so far. For the first time, development of real-time PCR assay for chestnut allergen in processed foods described. Initially, three sets novel primers allergen-encoding genes (Cas s 5, Cas 9 and TLP) were designed evaluated detection. Primers TaqMan probe finally selected sensitive binary mixtures with practical limit (LOD) 100 ppm. Effect processing sensitivity slightly reduced after boiling treatment. Amplification was importantly affected when heat pressure applied (autoclaving). Applicability tested by analysis commercial products. Consequently, results support validation accurate labelling.
