Ondansetron and sertraline may interact with 5-HTTLPR and DRD4 polymorphisms to reduce drinking in non-treatment seeking alcohol-dependent women: exploratory findings.

摘要: Abstract The purpose of this exploratory study was to examine the interaction 5-HTTLPR and DRD4 exon III polymorphisms with gender in non-treatment seeking alcohol-dependent (AD) individuals while alternately taking ondansetron sertraline. Evidence suggests that alcohol dependence may be influenced by a genetic gender-specific temporal changes making pharmacological treatment serotonergic drugs complex. main trial within-subject double-blind placebo-controlled human laboratory 77 non-treatment-seeking AD randomized (55 completed, 49 complete data) receive 200 mg/day sertraline or 0.5 mg/day for 3 weeks followed an self-administration experiment (ASAE), then placebo second ASAE, alternate drug, counterbalanced order, third ASAE. Results men were not significant. Women LL genotype receiving SS/SL (matched), drank significantly fewer drinks per drinking day (DDD) during 7 days prior first ASAEs than women mismatched medication (i.e., SS/SL). In 3-way interaction, alleles medications, who received had ≥7 repeats DDD as did but conversely ad libitum these same ASAEs. These findings add possible support differences among response pharmacotherapies. Future trials should powerful enough take into account endophenotypes targeting interactions essential successfully treat dependence.
