Adipose Triglyceride Lipase and Hormone-sensitive Lipase Are the Major Enzymes in Adipose Tissue Triacylglycerol Catabolism

摘要: The mobilization of free fatty acids from adipose triacylglycerol (TG) stores requires the activities lipases. In this study, we demonstrate that triglyceride lipase (ATGL) and hormone-sensitive (HSL) are major enzymes contributing to TG breakdown in vitro assays organ cultures murine white tissue (WAT). To differentiate between ATGL- HSL-specific cytosolic preparations WAT determine relative contribution these hydrolases lipolytic catabolism fat, mutant mouse models lacking ATGL or HSL a mono-specific, small molecule inhibitor for (76-0079) were used. We show 76-0079 had no effect on HSL-deficient but, contrast, essentially abolished acid ATGL-deficient fat. CGI-58, recently identified coactivator ATGL, stimulates hydrolase activity wild-type but not WAT, suggesting is sole target CGI-58-mediated activation lipolysis.
