Buyang Huanwu Decoction attenuates H2O2-induced apoptosis by inhibiting reactive oxygen species-mediated mitochondrial dysfunction pathway in human umbilical vein endothelial cells.

摘要: Apoptosis of endothelial cells caused by reactive oxygen species plays an important role in ischemia/reperfusion injury after cerebral infarction. Buyang Huanwu Decoction (BYHWD) has been used to treat stroke and stroke-induced disability, however, the mechanism for this treatment remains unknown. In study, we investigated whether BYHWD can protect human umbilical vein (HUVECs) from H2O2-induced apoptosis explored underlying mechanisms. To investigate effect on HUVECs, established a oxidative stress model detected Hoechst 33342 propidium iodide staining. JC-1 DCFH-DA assays,western blotting electron microscopy were examine apoptosis. Pretreatment with significantly inhibited protein caspase-3 expression concentration-dependent manner. addition, reduced production promoted endogenous antioxidant defenses. Furthermore, loss mitochondrial membrane potential structural disruption mitochondria both rescued BYHWD. protects HUVECs inhibiting damage dysfunction. These findings indicate that is promising ischemia diseases.
