摘要: Electronic nicotine delivery systems, or e-cigarettes, utilize a liquid solution that normally contains propylene glycol (PG) and vegetable glycerin (VG) to generate vapor act as carrier for flavorings. Evidence indicated these "carriers" reduced growth survival of epithelial cells including those the airway. We hypothesized 3% PG mixed with VG (3% PG/VG, 55:45) inhibited glucose uptake in human airway first step reducing cell survival. Exposure H441 bronchiolar (HBECs) PG/VG (30-60 min) mitochondrial ATP synthesis. glycolysis. mannitol volume height air-liquid interface cultures. Mannitol, but not increased phosphorylation p38 MAPK. transepithelial electrical resistance, which was associated solute permeability. decreased fluorescence recovery after photobleaching green fluorescent protein-linked transporters GLUT1 GLUT10, indicating transport function compromised. Puffing onto apical surface primary HBECs 10 min mimic effect e-cigarette smoking also transport. In conclusion, short-term exposure key components metabolism cells. propose this result membrane fluidity, further consequences on barrier function. Taking results together, we suggest factors contribute defensive properties epithelium. repeated/chronic agents are likely damage users.
