摘要: We describe novel transposon piggyBac vectors engineered to deliver transgenes as efficiently currently available systems, but with significantly less helper DNA co-delivered into the host genome. To generate these plasmids, we identified a previously unreported aspect of biology, that full-length terminal domains required for successful plasmid-to-chromatin transgene delivery can be removed from cassette other parts plasmid without impairing transposition efficiency. This is achieved by including in same plasmid, an additional sequence contains both long domains, modified prevent its design decreases size within delivered gene vector about 1500 just 98 base pairs. By removing sequences cassette, they are no longer incorporated genome which may reduce risk target cell transformation.
