摘要: When polyphenoloxidase (PPO) was exposed to sulfite prior susbstrate addition, inhibition irreversible. Trials regenerate PPO activity, using extensive dialysis, column chromatography, and addition of copper salts were not successful. Increased concentrations pH levels less than 5 enhanced the by sulfite. At 4, greater 0.04 mg/mL completely inhibited 1,000 units activity almost instantaneously. This suggested that HSO3- molecule main component in system inhibiting PPO. Column gel electrophoresis did demonstrate 35SO2 bound purified pear protein. Formation extra protein bands fractions on demonstrated. other evidence major mode direct irreversible modification structure, with retention its molecular unity.
