摘要: Objectives This study was to investigate the effect and mechanism of low-intensity pulsed ultrasound (LIPUS) on proliferation human amnion-derived mesenchymal stem cells (hAD-MSCs). Methods Human were isolated from amnion term placentas identified by flow cytometry differentiation culture. Proliferation hAD-MSCs investigated Cell Counting Kit-8, cell cycle EdU assays. Western blotting used determine protein expression levels. Results Human successfully as multipotent cells. Low-intensity promoted hAD-MSCs. analysis showed that LIPUS enter S G2/M phases G0/G1 phase. blot results phosphorylation activation ERK1/2 Akt significantly upregulated cyclin D1, E1, A2 B1. inhibitor (U0126) PI3K (LY294002) reduced LIPUS-induced Akt, respectively, which in turn hAD-MSCs. Conclusions Low-intensity can promote hAD-MSCs, PI3K-Akt signalling pathways may play important roles this process.
