摘要: Tumorigenesis is a multi-step process in which cells acquire capabilities that allow their growth, survival, and dissemination under hostile conditions. Different tests seek to identify quantify these hallmarks of cancerous cells; however, they often focus on single aspect cellular transformation and, fact, multiple are required for proper characterization. The purpose this work provide researchers with set tools assess vitro from broad perspective, thereby making it possible draw sound conclusions. A sustained proliferative signaling activation the major feature tumoral tissues can be easily monitored conditions by calculating number population doublings achieved over time. Besides, growth 3D cultures allows interaction surrounding cells, resembling what occurs vivo. This enables evaluation aggregation together immunofluorescent labeling distinctive markers, obtain information another relevant transformation: loss organization. Another remarkable characteristic transformed capacity grow without attachment other extracellular matrix, evaluated anchorage assay. Detailed experimental procedures evaluate cell rate, perform lineage markers cultures, test anchorage-independent soft agar provided. These methodologies optimized Breast Primary Epithelial Cells (BPEC) due its relevance breast cancer; applied types after some adjustments.
