摘要: Reversible myocardial dysfunction associated with transient ischemia has been termed the stunned myocardium. Because exogenous adenosine shown to protect ischemic myocardium, we hypothesized that augmentation of endogenous levels would attenuate stunning. To induce stunning, anesthetized dogs were subjected 15 minutes (left anterior descending artery occlusion) followed by 60 reperfusion. Erythro-9-(2-hydroxy-3-nonyl) adenine (EHNA; 5 mg/kg/hr), an deaminase inhibitor, was used augment levels. The effect EHNA on interstitial fluid (ISF) levels, coronary blood flow, and regional systolic wall thickening compared untreated group (n = 8). increased preischemia ISF threefold a corresponding increase in flow. administration did not alter thickening. Although fourfold during group, nearly sixtyfold above values EHNA-treated remained elevated throughout Postischemic function enhanced significantly treated EHNA. These data show inhibition attenuated Metabolic manipulation nucleoside may be beneficial limiting postischemic dysfunction.
