High-level expression of tuberous root storage protein genes of sweet potato in stems of plantlets grown in vitro on sucrose medium.

摘要: Sporamin, the tuberous root storage protein of sweet potato, accounts for about 60 to 80% total soluble this organ. The amount sporamin present in other organs is very low, or even not detectable, normal field-grown plants. However, stem potato plantlets grown axenically on agar medium containing sucrose was found accumulate large amounts sporamin. Two-dimensional gel electrophoretic profiles precursors synthesized vitro by poly(A)+ RNA are indistinguishable between roots plants and stems cultured plants, suggesting that an essentially identical set members multigene family expressed these two under different growth conditions. Transgenic tobacco having a CAT (chloramphenicol acetyltransferase) fusion gene with 5′ upstream region A gene, gSPO-A1, show preferential expression activity when maintained axenic culture as case potato. Deletion analysis revealed DNA sequence gSPO-A1 −94 −305, relative transcription start site, important its tobacco. This contains previously postulated putative regulatory elements conserved B genes.
