Antitumoral Alkylphospholipids Induce Cholesterol Efflux from the Plasma Membrane in HepG2 Cells

摘要: Alkylphospholipid (APL) analogs are promising candidates in the search for treatments of cancer. Previous studies conducted our laboratory indicate that, after prolonged treatment, they alter cholesterol homeostasis HepG2 cells. Here we describe effects that different APLs exert upon this cell line a 1-h exposure serum-free medium, including 1) rapid, significant increase efflux into extracellular which consequently provoked depletion plasma membrane (further assays an attempt to return control levels were only partially successful); 2) use methyl-β-cyclodextrin, indicated acted way similar agent is used frequently modulate levels; 3) phosphorylation Akt showed critical regulator survival was modulated by changes induced APLs; and 4) not related impairment traffic produced APLs.
