摘要: Abstract Background Immunohistochemical staining of entire nerve fibres allows for studying the molecular composition functional fibre subunits and may add to diagnostic value teasing. New method In this study, we established a sealed-slide reproducible immunostaining deep axoplasmic proteins in permanently straightened fibres. Results Immunostaining teased very much is facilitated by tip-fixation with biocompatible glass adhesives. Antibody penetration fresh nerves can be achieved thermic chemical permeabilisation while enzymatic digestion sufficient permeability after aldehyde fixation. Comparison existing methods The recommended herein are easy perform represent reliable way whole mount immunostaining. Conclusions Sealed-slide tip-fixed permeabilised biopsies will help validate neurophysiological abnormalities screen target molecules predictive markers peripheral disorders such as inherited neuropathies Guillain-Barre syndrome.
