摘要: Publisher Summary This chapter presents a procedure for the preparation of catechol O-methyltransferase that catalyzes transfer methyl group from S-adenosylmethionine (AdoMet) to substrate, resulting in formation meta- and para-O-methylated products. The convenient assay activity is radiochemical technique using either [14CH3]AdoMet or [C3H31AdoMet. 14CH3- C3H3-labeled O-methylated products are isolated by simple solvent extraction. based on use [14C]AdoMet 3,4–dihydroxybenzoic acid as substrates, followed extraction 14CHh-labeled (3-hydroxy-4-methoxybenzoic 4-hydroxy-3-methoxybenzoic acid).
