摘要: Background Massively parallel pyrosequencing of amplicons from the V6 hypervariable regions small-subunit (SSU) ribosomal RNA (rRNA) genes is commonly used to assess diversity and richness in bacterial archaeal populations. Recent advances technology provide read lengths up 240 nucleotides. Amplicon can now be applied longer variable SSU rRNA gene including V9 region eukaryotes. Methodology/Principal Findings We present a protocol for amplicon eukaryotic environmental samples biodiversity inventories species estimation. The International Census Marine Microbes (ICoMM) Microbial Inventory Research Across Diverse Aquatic Long Term Ecological Sites (MIRADA-LTERs) projects are already employing this tag sequencing wide both marine freshwater environments. Conclusions/Significance provides means estimating deeply-sampled populations discovering novel environment.
