New method for culture of zona-included or zona-free embryos: the Well of the Well (WOW) system.

摘要: Culture of mammalian zygotes individually and in small groups results lower developmental rates than culture large groups. Zona-free also have impaired potential current systems. This paper describes a new approach to resolve the problems, Well (WOW) system. Small wells (WOWs) were formed four-well dishes by melting bottom with heated steel rods. The WOWs then rinsed, filled medium, embryos placed into WOWs. To test value WOW system 3 × factorial experiment was performed. Bovine presumptive cultured from day 1 7 (day 0: insemination) using three modules (single embryos, embryo five, or single zona-digested embryos) different systems (400 μl 200 drops, WOWs). An additional control group consisted 40 50 400 medium. resulted higher blastocyst/oocyte for all (single: 59%; five: 61%; zona-digested: 53%) drops (P < 0.05 all). rate independent number per well. cell blastocysts did not differ that controls.
