摘要: Chromatin, the physiological template for transcription, plays important roles in gene regulation by nuclear receptors (NRs). It can (1) restrict binding of NRs or transcriptional machinery to their genomic targets, (2) serve as a target regulatory posttranslational modifications NR coregulator proteins with histone-directed enzymatic activities, and (3) function scaffold variety transcription-related proteins. The advent vitro "cell-free" systems that accurately recapitulate ligand-dependent transcription chromatin templates has allowed detailed analyses these processes. Biochemical studies have advanced our understanding mechanisms regulation, including role ligands, coregulators, nucleosome remodeling. In addition, they provided new insights about dynamics NR-mediated transcription. This chapter reviews current methodologies assembling, transcribing, analyzing vitro, well information been gained from studies.
