Opening of Compacted Chromatin by Early Developmental Transcription Factors HNF3 (FoxA) and GATA-4

摘要: The transcription factors HNF3 (FoxA) and GATA-4 are the earliest known to bind albumin gene enhancer in liver precursor cells embryos. To understand how they access sites silent chromatin, we assembled nucleosome arrays containing sequences compacted them with linker histone. GATA-4, but not NF-1, C/EBP, GAL4-AH, bound their chromatin opened local nucleosomal domain absence of ATP-dependent enzymes. ability open is mediated by a high affinity DNA binding site C-terminal protein, which binds histones H3 H4. Thus, that potentiate development inherently capable initiating opening events.
