摘要: Expression of the human osteocalcin promoter is negatively regulated by glucocorticoids in vivo. In vitro DNase I and exonuclease III footprinting analysis showed binding purified glucocorticoid receptor close proximity to overlapping with TATA box gene. These results imply competition or interference box-binding transcription factor IID as a mechanism repression this gene glucocorticoids. support notion, point mutation site indicated that flanking nucleotides not motif per se were important for interaction. Moreover, DNA assays specific only region corresponding an immunoglobulin heavy-chain promoter.
