摘要: Abstract The Fusarium oxysporum gene nia , encoding nitrate reductase (NR), was isolated from a cosmid library by direct complementation of an F. − mutant. specifies protein 905 amino acids and contains 57-bp intron. Comparison the deduced aa sequence with NR other fungi revealed high degree similarity conservation in intron position. cloned made it possible to develop first homologous transformation system for this fungus. Transformation frequencies up 600 transformants per μg DNA were achieved. Gene replacements, single-copy integrations at non-homologous sites observed. Direct comparison between plasmids cosmids carrying same showed higher frequency targeted using vectors. replacement events observed about 50% analysed each type vector used. This substitution offers new applications .
