Fyn mediates transforming growth factor-beta1-induced down-regulation of E-cadherin in human A549 lung cancer cells.

摘要: Transforming growth factor-beta (TGF-β) signaling positively contributes to the regulation of tumor metastasis. However, underlying molecular mechanisms are less well defined. We here show that Fyn, a member Src family tyrosine kinases, plays critical role in mediating TGF-β1-induced down-regulation E-cadherin human A549 lung cancer cells. Blockade Fyn with siRNA knockdown or ligand-binding defective mutant significantly lowered ability TGF-β1 repress expression. Furthermore, our results demonstrated facilitates TGF-β1-mediated suppression through p38 kinase-dependent induction Snail. Collectively, findings identify Fyn-p38-Snail cascade as new pathway oncogenic TGF-β function.
