摘要: Single molecule fluorescence techniques offer unique insights into mechanisms of conformational changes RNA. Knowing how to make fluorescently labeled RNA molecules and understanding potential limitations different labeling strategies is essential for successful implementation single techniques. This chapter offers a step by overview the process obtaining constructs ready measurements. Several alternative methods are described each step, ways troubleshooting most common problems, in particular, splinted ligation, suggested.
