摘要: Abstract A large production of leukotrienes (LTs) can be induced in human eosinophils or neutrophils by priming with granulocyte-macrophage colony-stimulating factor and subsequent stimulation platelet-activating (PAF) the anaphylatoxin C5a. Here, we investigated effects a plant extract petasites hybridus (Ze339) its isolated active sesquiterpene ester petasin these two vitro cell models. Zileuton, 5-lipoxygenase inhibitor, was used as positive control. All compounds inhibited both cysteinyl-LT synthesis LTB 4 neutrophils. In contrast, only Ze339 petasin, but not zileuton, abrogated PAF- C5a-induced increases intracellular calcium concentrations. These data suggest that may block, compared to earlier signalling events initiated G protein-coupled receptors granulocytes, perhaps at level proximal phospholipase C β . Taken together, appears one major compound extract, since it demonstrates same inhibitory activities on fluxes LT generation does.
