Role of Acinus in regulating retinoic acid-responsive gene pre-mRNA splicing.

摘要: Acinus-S’ is a corepressor for retinoic acid receptor (RAR)-dependent gene transcription and has been suggested to be involved in RNA processing. In this study, the role of Acinus isoforms regulating pre-mRNA splicing was explored using vivo assays. Both Acinus-L Acinus-S’, with activity higher than that increase (RA)-responsive minigene containing weak 5′ splice site but not RA-responsive strong site. RA treatment further enhances by dose- time-dependent manner, suggesting RA-dependent addition RA-independent Acinus. The effect occurs varying degrees constructs several different promoters, while specific transcripts derived from driven response element (RARE)-containing promoter. This suggests ligand-dependent related RA-activated RAR bound RARE. RRM domain necessary repressed RNPS1. Importantly, measurement endogenous human RARβ Bcl-x demonstrates stimulates use weaker alternative these two genes manner Bcl-x.
