Efficient large-scale development of microsatellites for marker and mapping applications in Brassica crop species.

摘要: A set of 398 simple sequence repeat markers (SSRs) have been developed and characterised for use with genetic studies Brassica species. Small-insert (250-900 bp) genomic libraries from rapa, B. nigra, oleracea napus, highly enriched dinucleotide trinucleotide SSR motifs, were constructed. Screening the clones a mixture oligonucleotide probes revealed positive hybridisation to between 75% 90% clones. Of these, 1,230 sequenced. Primer pairs designed clones, 270 (67.8%) amplified PCR product expected size in their focal and/or closely related further screen 138 primers that produced napus germplasm found 86 (62.3%) length polymorphisms within at least one line test array representing four The results this used identify 56 SSRs combined 41 had previously shown polymorphism parents mapping population.
