Inhibitors of Anion Exchanger Activity Reduce Sodium Chloride-Dependent Taurine Transport by Brush Border Vesicles

摘要: The erythrocyte anion exchanger, or band 3 protein, catalyzes the exchange of chloride and bicarbonate in many cells. Anion exchanger activity is predominantly found basolateral membrane cortical collecting duct. Because proximal tubular renal taurine uptake across brush border surface dependent upon sodium chloride, effect inhibitors on accumulation by rat vesicles was examined. probes, DIDS (4,4-’diisothiosulfonic acid), NBD-taurine (N-[7-nitrobenz-2-oxa-1,3-diazol-4-yl]-aminoethane sulfonic were used as activity. DIDS, NAP-, all markedly inhibit initial rate NaCl-dependent vesicles. NAP- because inhibition not when performed presence NaNO3 place chloride. In maximal NBD-taurine, no additional evident after incubation with DIDS. On other hand, a competitive inhibitor uptake, β-alanine, used, (p < .01 respectively). These results suggest some interaction process at apical tubule. fluorescent probe, it may be possible to isolate peptides that bind demonstrate fluorescence. Preliminary isoelectric focusing experiments protein incubated show fluorescence localized several particular fractions.
