摘要: The enzyme 8-oxo-7,8-dihydrodeoxyguanosine triphosphatase (8-oxo-dGTPase) hydrolyzes 8-oxo-dGTP to 8-oxo-dGMP, thereby preventing misincorporation of into DNA. We investigated expression MTH1 gene encoding 8-oxo-dGTPase. Large amounts mRNA were present in thymus and testis, embryonic tissues, certain cell lines. In peripheral blood lymphocytes, the level was significantly increased after concomitant treatment with phytohemagglutinin interleukin-2. Analyses 5' regions transcripts revealed that 7 types mRNAs, which may be produced by transcription initiation at different sites and/or alternative splicing. consists 5 major exons, some are composed differentially processed segments. All mRNAs carry entire coding region, functional. Three ATG codons in-frame found mRNAs. There is a polymorphic alteration splicing site (GT GC) located exon 2, an event affects patterns transcript. Allele frequency this polymorphism about 20% among healthy volunteers.
