摘要: Abstract The cytotoxic effects of conventional (doxorubicin, 5-fluorouracil, cisplatin) and investigational (2′,2′-difluorodeoxycytidine, hexadecylphosphocholine, EO9, rhizoxin) anticancer drugs were studied in subconfluent multilayered postconfluent cultures human colon ovarian carcinoma cell lines. Chemosensitivity was assessed 4 days after a 24-h drug exposure with the sulphorhodamine B assay. Except for rhizoxin, all tested yielded an ec 50 (drug concentration producing absorbance readings 50% lower than those non-treated wells) that higher obtained cultures. Compared cultures, showed decreased cellular nucleotide concentrations ATP/ADP ratios, addition to increased percentage G 0 /G 1 activity DT-diaphorase, reductase involved bioactivation similar sub- These results indicate similarity model presented vivo models more complex vitro techniques.
