摘要: We present the results of studies an aqueous sample a highly {15N,2H} enriched protein, SH3 domain from Fyn. Measurements 1H relaxation and interactions between H2O solvent exchangeable protons are given, as well method for increasing effective longitudinal proton resonances. The long-range isotope shifts measured, 15N, which arise due to perdeuteration. Simulations, employed 7 or 8 spin matrix analysis, were compared experimental data time series 2D NOESY datasets some agreement experiment simulation suggest that, with this dilute sample, relatively long mixing times (up 1.2 s) can be used detect specific dipolar amide up about 7A apart. A set 155 inter-amide NOEs side chain thus identified in 3D HSQC-NOESY-HSQC experiments. These data, alone combination previously collected restraints, calculate sets structures using X-PLOR. available X-ray NMR Fyn domain.
