Fabrication of Protein Microarrays for Immunoassay Using the Electrospray Deposition (ESD) Method

摘要: In recent years new methods for active protein film fabrication have been rapidly developing. One of these is the electrospray deposition (ESD) method. This method allows spontaneous many identical dots and has a remarkable spatial resolution as well ability to overcome limitations like low rates deposition, efficiency substance transfer, cross-contamination. this study, an antibody-based array high-throughput immunoassay was fabricated with ESD using quartz mask holes made by abrasive jet technique. An antibody solution electrosprayed onto ITO glass then deposited antibodies were cross-linked vapor glutaraldehyde. The diameter spots approximately 150 μm. arrays incubated corresponding target antigenic molecules washed. captured antigens collectively detected fluorescence chemiluminescence. These signals quantitatively visualized high-resolution charge-coupled device (CCD) detector. Sensitive (~1 ng/ml) simultaneous detection various could be performed enzyme linked immunosorbent assay or system.
