摘要: DNA excision repair plays a significant part in platinum-based chemotherapy by removing lesions caused platinum-containing drugs. The nucleotide (NER) pathway is the mammalian mechanism that removes bulky adducts induced damaging chemotherapeutic agents. Platinum compounds induce their cytotoxic effect binding to molecule form of platinum-DNA-adduct. NER main responsible for platinum resistance increased platinum-DNA-adduct removal and cross complementing-group 1 (ERCC1) gene major role NER-pathway because its damage recognition ability. This chapter will review mechanisms as it relates regulation ERCC1. A brief discussion on cancer stem cells also presented.
