Antifungal activity of spider venom-derived peptide lycosin-I against Candida tropicalis.

摘要: Abstract Candida species are a major cause of human mucosal and deep tissue fungal infections, but few antifungal treatments available. Here, we showed that lycosin-I, peptide isolated from venom the spider Lycosa singoriensis, acted as potent inhibitor against species. The MIC50 values lycosin-I reached 8 μg/mL to treat fluconazole-susceptible fluconazole-resistant C. tropicalis isolates. Time-kill kinetics assays revealed after 2-hour exposure, reduced colony-forming units/mL in isolates approximately 70%. Furthermore, salinity tolerance suggested even presence Mg2+, maintained its ability at high concentration. When concentration was increased 1 × MIC 8 × MIC, significant decrease biofilm metabolic activity observed both Moreover, inhibitory 50 (BIC50) eradicating (BEC50) were 32 μg/mL 128 μg/mL, respectively. Confocal laser scanning microscopy localization CY5-labeled mainly cell cytoplasm, likely be localized cytoplasm transportation across wall membrane. Overall, our work shows is agent with efficacy, tolerance, anti-biofilm properties.
