摘要: We have developed a microdissection technique that allows for procurement and analysis of specific, minute cell populations from routine, 5-mu, formalin-fixed, paraffin-embedded histological tissue sections. Lesions < 1 mm in size can be specifically examined. Cells interest are procured under direct microscopic visualization followed by single-step DNA extraction subsequent polymerase chain reaction. Amplification selected was demonstrated detecting loss heterozygosity (LOH) at the von Hippel-Lindau disease (VHL) gene an atypical renal lesion carcinoma kidney VHL patient. Moreover, previously unrecognized LOH on short arm chromosome 3 (3p25-26) detected microdissected colorectal cells non-VHL patient with sporadic colon carcinoma.
