摘要: A new protocol for the purification of phytochrome from etiolated Avena sativa L. seedlings in 15-25% yield is described. This preparation exhibits molecular properties dissimilar to those previously reported oats. Phytochrome prepared by this method contains a dominant species having Mr = 124,000 (70% total) on sodium dodecyl sulfate-polyacrylamide gels, addition chromoprotein with 118,000 (30%). blocked NH2 terminus indicated amino acid sequencing purified preparation. By ultraviolet-visible spectroscopy, displays 730-nm absorption maximum Pfr, an enhanced APfr730/APfr668 ratio under saturating red light illumination, and reduced amount dark reversion. Circular dichroism reveals band Pfr blue spectral region. These electrophoretic behavior are consistent available information vivo or rapidly extracts oat seedlings.
