摘要: Florida panthers are endangered pumas that currently persist in reduced patches of habitat South Florida, USA. We performed mitogenome reference-based assemblies for most parental lines the admixed resulted from introduction female Texas into 1995. With addition 2 puma mitogenomes, we characterized 174 single nucleotide polymorphisms (SNPs) across 12 individuals. defined 5 haplotypes (Pco1-Pco5), one which (Pco1) had a geographic origin exclusive to Costa Rica and Panama was possibly introduced Everglades National Park, prior Haplotype Pco2 native Florida. Haplotypes Pco3 Pco4 were Texas, whereas haplotype Pco5 an undetermined origin. Phylogenetic inference suggests Pco1-Pco4 diverged ~202000 (95% HPDI = 83000-345000) years ago Pco2-Pco4 ~61000 9000-127000) ago. These results congruent with south-to-north continental expansion recent North American colonization by pumas. Furthermore, may have migrated no earlier than ~44000 2000-98000) Synonymous mutations presented greater mean substitution rate other mitochondrial functional regions: nonsynonymous mutations, tRNAs, rRNAs, control region. Similarly, all protein-coding genes under predominant negative selection constraints. directly indirectly assessed presence potential deleterious SNPs ND2 ND5 as consequence Screenings such variants recommended extant panthers.
