Tumor necrosis factor receptor p75 mediates cell-specific activation of nuclear factor kappa B and induction of human cytomegalovirus enhancer.

摘要: The functional role of human tumor necrosis factor receptor (TNFR) p75 was studied by the use TNFR p75-specific agonistic antibodies. Human SW480T adenocarcinoma cells, stably transfected with a reporter construct containing beta-galactosidase under control cytomegalovirus immediate early enhancer, were stimulated anti-TNFR polyclonal antiserum or monoclonal antibodies followed measurement activity and analysis electrophoretic mobility shift assays. It found that cross-linking led to strong induction enhancer as well activation nuclear factor-kappa B (NF-kappa B). Stimulation also mediated NF-kappa in KYM-1 rhabdomyosarcoma cells but not other cell types such U937 HL-60 monocytic Eahy 926 endothelial cells. induced delayed approximately 15 min compared p55, indicating two TNFRs activate through different signaling pathways. data presented this study identify intracellular responses which have been reported previously suggest p75-induced is strictly type-specific.
