摘要: A method based on the Competitive Index was used to identify Salmonella typhimurium virulence gene interactions during systemic infections of mice. Analysis mixed involving single and double mutant strains showed that OmpR, type III secretion system pathogenicity island 2 (SPI-2) SifA [required for formation in epithelial cells lysosomal glycoprotein (lgp)-containing structures, termed Sifs] are all involved same function. sifA expression induced after entry into host dependent SPI-2 regulator ssrA. sifA(-) strain had a replication defect macrophages, similar ompR(-) strains. Whereas wild-type reside vacuoles progressively acquire lgps vacuolar ATPase, majority bacteria lost their membrane were released cell cytosol. We propose strain, through action effectors (including SpiC), diverts Salmonella-containing vacuole from endocytic pathway, subsequent recruitment maintenance ATPase/lgp-containing membranes enclose replicating is mediated by translocation SifA.
