Icariin protects murine chondrocytes from lipopolysaccharide-induced inflammatory responses and extracellular matrix degradation.

摘要: Abstract Septic arthritis is an inflammatory arthropathy characterized by degeneration of articular cartilage. Icariin, the main active flavonoid glucoside isolated from Epimedium pubescens , used as antirheumatics (or antiinflammatory), tonics, and aphrodisiacs in traditional Chinese medicine. In this study, we lipopolysaccharide (LPS) to simulate vitro response chondrocytes during septic arthritis. Our hypothesis that icariin can protect LPS-induced inflammation extracellular matrix degradation. The neonatal mice was induced LPS antiinflammatory effects were examined. synthesis nitric oxide analyzed, whereas titer glycosaminoglycan total collagen measured gene expressions (including inducible synthase [iNOS], metalloproteinase [MMP]–1, MMP-3, MMP-13) evaluated. results showed viability chondrocytes, synthesis, significantly decreased, increased presence 10 −5 g/mL LPS. Icariin pretreatment partially reverse these effects. up-regulated MMP-1, 3, 13, cyclooxygenase-2 (COX-2), iNOS genes treatment also down-regulated 1.8%, 0.056%, 7.7%, 3.1%, 5.3% LPS-positive control sample, respectively. demonstrate a safe anabolic agent chondrocytes. may exert its protective through inhibition MMP then reduce destruction.
