摘要: Epidemiological data have indicated that estrogen replacement therapy (ERT) can decrease the risk of developing Alzheimer's disease (AD). Phytoestrogens been proposed as potential alternatives to ERT. The aim present study was assess neuroprotective effects puerarin, a phytoestrogen isolated from Pueraria lobata, against toxicity beta-amyloid (Aβ) in relation mitochondria-mediated cell death process, and elucidate role activation Akt modulation pro- antiapoptotic proteins puerarin-induced neuroprotection. shows puerarin afforded protection Aβ-induced through inhibiting apoptosis PC12 cells. This result also confirmed by activated caspase-3 assay. P-Akt, Bcl-2 p-Bad expression increased after pretreatment with cells exposed Aβ(25-35), whereas Bax cytochrome c release decreased. Interestingly, these Aβ(25-35) insult were abolished wortmannin, an inhibitor PI3K phosphorylation. These findings suggest prevent neurotoxicity neuronal apoptosis, might be preventive or therapeutic agent for AD.
