摘要: Abstract Using drugs in combination can be important therapeutic strategies, both to decrease the risk of toxic side effects as well likelihood resistance evolving. These properties are largely affected by interactions among drugs. Given increased use three or more clinics, we provide a review challenges researchers face when studying higher-order drug together with current theoretical and methodological advances resolving these issues. The include deriving measures for interaction compared single efficacies, disentangling emergent from lower-order interactions, enhancing resolution data better classify types, considering practical difficulties such measuring responses combinations across range concentrations. systems biology modeling approaches reviewed here offer ways go beyond-pairwise quantify understand interactions. However, there many research directions yet pursue mechanistic insights determine consequences
